Treatment for glioblastoma (GBM) patients is surgical resection of the tumor, followed by radiation and chemotherapy with temozolomide (TMZ). Unfortunately, 60% of newly diagnosed GBM patients express high levels of the DNA repair enzyme O6-methylguanine-DNA methyltransferase (MGMT) and are resistant to TMZ, and all patients eventually become refusing treatment. The blood-brain barrier (BBB) is a barrier to the delivery of chemotherapeutic agents in GBM, and BBB-permeable agents are effective in TMZ-resistant patients and those who have significant relapses. Large amino acid transporter 1 (LAT1) is expressed at the BBB and in GBM, it is detected at low levels in normal brain tissue. A selective LAT1 therapy would be able to target brain tumors while avoiding taking over healthy tissue.
We report a new chemical compound (QBS10072S) combining a potent cytotoxic chemotherapeutic domain (tertiary N-bis(2-chloroethyl) amine) and structural features of a selected LAT1 substrate and tested it against GBM models.
