By using microfluidic particles to incorporate cell-free activity into inert hydrogel beads, we have developed a platform that can perform biologically relevant functions without the need for cells. Specifically, cell-free lysates serve a benefit in performing cellular functions and providing biologically relevant metabolic products without the need for optimal biological conditions for cell growth and proliferation. By triggering specific biological components that allow transcription and translation to occur, these cell-like functions can be remodeled in vitro without the need for the whole cell and organelle environment. This enables the optimization of synthetic biological circuits, either via focus or logical switches, simply by adding or removing genetic components (plasmids, inducers, or repressors) of the regulatory elements. Here, we show an application of robust, portable, substrate-independent, cell-free processes to advance the sensing and reporting functions of the quorum-sensing molecule N-3-oxododecanoyl homoserine lactone (3OC12HSL) found critical for pathological Pseudomonas aeruginosa infection. We are developing an easily adaptable agarose bead platform that can be simply programmed to suit a variety of biological and chemical sensing applications for easy delivery and activation in remote environments – even in conditions with very little wetting.

Recent innovations using cell-free lysates have led to a variety of applications from biocomputing [6, 7] molecular design and engineering [8, 9], engineering of metabolic pathways [10, 11, 12] to new approaches to protein synthesis [13, 14]. , 15th]. In the case of E. coli, the coupled transcription and translation machinery enables “one vessel” protein synthesis, but is limited by types of complex proteins such as post-translational modified proteins as well as mixed proteins such as lipoproteins and proteins with large portions of the carbohydrate fraction as mostly found in cells the reality of muclear. While cell-free lysates from rabbit reticulocytes enable full-length, post-translational modified proteins, protein yields are less desirable due to the presence of other endonucleases and RNA that inhibit efficient translation [14]. Compared with wheat germ-based cell-free lysates, exotic RNAs such as double-stranded RNAs and proteins with heavy thiol moieties are translated very efficiently, but this cell-free lysate requires 5 cap modifications on the mRNAs [15]. In addition to uses for bulk protein synthesis, cell-free lysates are also the basis for precise input-output logic functions [5, 21]. Several groups have explored the use of these components for disease diagnosis and detection of small molecules and environmental hazards [22]. Specifically, by combining the use of cell-free analyzers with paper microfluidics, cell-free solutions provide the detection device component for strain-specific pathogens such as Ebola virus as well as environmental toxins such as arsenic [23, 24, 25]. However, due to the volume of samples needed to perform cell-free paper diagnoses and the rates of false positive detection, these devices may not be effective or efficient in detecting pathogens and toxins in real time. Limited volumes are especially relevant when dealing with blood as an analyzer. Others have improved the reliability and sensitivity of these devices using high-throughput microfluidics, but these also require the use of volumes greater than 100 μl of analytes [26, 27]. One possible answer to the size problem is to reduce the reaction size and take advantage of the picomolar sensitivity (0.001 nm) of these lysates by encapsulating cell-free lysates in micro-engineered vessels that are semi-permeable to the environment.
